Modeling of the Effects of Threonine, Valine, Isoleucine and Pyridoxal 5'-monophosphate on Biosynthetic Threonine Dehydratase Reaction

چکیده
Motivation: Development of mathematical models that adequately describe moleculargenetic mechanisms is one of the main tasks in modern bioinformatics. Not only should a model be sensitive to the particulars of enzyme reactions underlying cell life activity, but also to regulation exerted on enzyme activity by metabolites and specialized cell proteins. Results: The gene network for branched-chain amino acid biosynthesis in E. coli has been reconstructed using the GeneNet system. On the basis of available information on this gene network, elementary mathematical models for enzyme reactions have been developed using generalized Hill functions. We herein give a detailed description of the mathematical model for the biosynthetic threonine dehydratase-catalyzed reaction that yields OBUT; the model includes enzyme activity regulation. Availability: The model is available on request. Gene network is available at http://wwwmgs.bionet.nsc.ru/mgs/gnw/genenet/viewer/index.shtml.

Cooperative binding of the feedback modifiers isoleucine and valine to biosynthetic threonine deaminase from Escherichia coli.
Control of the regulatory enzyme threonine deaminase from Escherichia coli is achieved by isoleucine inhibition and valine activation. The mechanism by which these heterotropic effectors regulate the enzyme was investigated by measuring the binding of isoleucine and valine by spectroscopic, kinetic, calorimetric and equilibrium dialysis techniques. The addition of isoleucine or valine to threon...
متن کاملPurification and feedback control of threonine deaminase activity of Rhodopseudomonas spheroides.
1. Threonine deaminase has been purified about 1400fold from extracts of the photosynthetic bacterium Rhodopseudomonas spheroides by ammonium sulfate fractionation and by chromatography on DEAE-cellulose and hydroxylapatite columns. The purified enzyme is almost completely resolved with respect to bound coenzyme, and the activity of holoenzyme is specific for pyridoxal phosphate. The enzyme is ...
متن کاملEnergetics of cooperative ligand binding to the active sites of biosynthetic threonine deaminase from Escherichia coli.
The sigmoidal kinetics of alpha-ketobutyrate production catalyzed by threonine deaminase are shifted in the presence of the feedback inhibitor isoleucine and the activator valine to control carbon flow through branched-chain amino acid biosynthesis in Escherichia coli. As an initial effort toward developing a molecular mechanism for cooperativity and feedback inhibition in this enzyme, the bind...
متن کاملHomocysteine toxicity in Escherichia coli is caused by a perturbation of branched-chain amino acid biosynthesis.
In Escherichia coli the sulfur-containing amino acid homocysteine (Hcy) is the last intermediate on the methionine biosynthetic pathway. Supplementation of a glucose-based minimal medium with Hcy at concentrations greater than 0.2 mM causes the growth of E. coli Frag1 to be inhibited. Supplementation of Hcy-treated cultures with combinations of branched-chain amino acids containing isoleucine o...
متن کاملEnzymes of the isoleucine-valine pathway in Acinetobacter.
Regulation of four of the enzymes required for isoleucine and valine biosynthesis in Acinetobacter was studied. A three- to fourfold derepression of acetohydroxyacid synthetase was routinely observed in two different wild-type strains when grown in minimal medium relative to cells grown in minimal medium supplemented with leucine, valine, and isoleucine. A similar degree of synthetase derepress...
